Exactly what it says on the tin...

Recently, but it makes sense. Girly means the character is feminine, ambiguous characters need to have either no masculine or feminine traits, or have conflicting traits.
These requirements conflict with each other, if an ambiguous character is girly they're not ambiguous by definition.

Ah, alright. Fair enough.

Heh. It was originally (pre-2013) only tagged for males, and I had hard time adapting to the 'new' "it's now also tagged for ambiguous" change.
Though.. girly and manly are now both taggable for cuntboy and male_herm, according to the wiki? Whereas female, dickgirl and non-male herm lack such tags. Not sure how that works.

Wait, sudden thought...Does this mean that pics tagged with both ambiguous and girly should be changed to female? 'cause that'd mean they have enough feminine traits somebody interpreted them as girly, with little to nothing in-image to contradict the female tag...

I always felt that tags like ambiguous_gender and girly shouldn't be able to both be tagged on the same picture because it is too contradictory. But at the same time, I also think that ambiguous characters should only be tagged with a sex if the author specifically states their sex (going against the "tag what you see" approach on the site) and ruling out the ambiguity.
For example; this is tagged as both girly and ambiguous, whereas I feel just an ambiguous tag should be there: post #1304270
To my knowledge, the character isn't assigned any sex, and they lack any discernible gender features. Therefore, they're just as girly as they are manly unless the author states otherwise.
Bottom line: gender tags can kind of be a mess, and everyone will have a different opinion on them.

Ambiguous_gender and girly are not measuring the same thing, though. They are measuring partly the same thing and partly a different thing.
Ambiguous_gender seems to be about physical characteristics (nothing else is explicitly mentioned) whereas girly is about "personality, clothing, and sometimes body type".
Even though the girly wiki says not to use girly for ambiguous_gender characters, the definitions themselves make it difficult to see how this makes sense - girly is neither entirely within the conceptual space that ambiguous_gender et al. want to quantify, nor entirely outside of it. Sharkish's example illustrates the problem well.

Wouldn't this ruling make it almost impossible for a safe image of a femboy to not be tagged female?

there is difference between character with masculine physique + feminine traits and character with completely ambiguous physique + feminine traits. genitals and breasts are not the only thing taken in account in gender tagging.

No I wasn't. What are you on about? I figured it went without saying that my question only applied to solo-character images...
