I'm wondering, what functions do ampersands actually cause on eSix searches and blacklists (like with hyphens to avoid a tag or metatag, or asterisks for wildcards)? I can't seem to find anything about them in the help.

The proper spelling for that copyright is Sam & Max, though.

Yeah, but unfortunately, the blacklist has issues with tags using ampersands. I tested this issue by adding puzzle_&_dragons to my blacklist and I could still see posts relating to that copyright when I search for it, only hiding posts that are tagged with other tags on my blacklist. For comparison, I added lady_and_the_tramp to my blacklist to test that tag out (since the copyright obviously isn't spelled with an ampersand), and all such posts are properly hidden as intended.
The OP brought up this ampersand issue at forum #269389.

Bumping this thread because the ampersand bug is still very much active and thus every tag that has & in it still cannot be hidden without resorting to using the percent-encoded form of the character (%26) for the blacklist to work with them. Most of the other tags with ampersands (primarily copyright and character tags) should also be aliased with their relevant aliases and implications moved.

This was something I was going to do eventually, and I've done it with a chunk of ampersand tags. However, obviously, there's still some I need to get to, and will get to it soon

Yep, Sam & Max bunch are done and approved, and currently I'm in the works of doing a good chunk of other ampersand tags
