e621 is the "E Number" (or the Europeon Union chemical code number) for Monosodium Glutamate which is a food additive that enhances flavor.

I think the reason was that this site enhances the internet just like monosodium glutamate enhances food. But I could be wrong about that reasoning. It was before my time.
