Allow me to clarify the above, when River first released, her bio described her as the "Calm sister to Thane". Since then her Bio has been changed and the no supplemental material exists to confirm or deny this. Thanes Novalis made River a twin sister to another wolf but it also does not deconfirm their relation.

One line of dialogue still exists and its Rivers win quote; "Me queen? Well I can't be any worse then Thane" which does heavily support that they're related/siblings.

Considering the devs like to equate Armello to a playable Game of Thrones, I doubt these two would be above Incest to obtain the throne.
