After a very brief test, it appears you can't blacklist user favorites. Just add "-fav:Ponry" to your search terms.
I do believe it's one of the features of re621 if you're willing/able to install a userscript though.
The default blacklist implementation supports the special tag fav:me and only fav:me

Yes. It will blacklist images that you have favorited.
