Artist here: They don't. They just have different sexual preferences, in addition to names and personality. They're both male.

I'm not sure which is funnier: Diglett being perfectly innocent and having no idea what he's delivering, or Diglett absolutely knows what he's delivering and being very cheerful about it.

Ok. I know you needed to do something to differentiate them, the problem is the eyelashes (or what appears to be eyelashes) are usually done in that manner for a female, so it was natural to believe they were female.
