Seems like it was originally used to refer to anime with excessive fan-service? I'm not finding any official definitions that use plot to actually refer to real equines, but I only put in 5min of effort to look.

The word Plot can mean specifically a horse's butt. Hence the pun in the picture as well as bronies who are interested in this sort of art using the pun frequently.

The use of "plot" for "horse ass" comes specifically from an early MLP meme with shots of pony asses and the caption "I watch it for the plot." It was not used earlier than that.
