Actually, it is confirmed that Nick's mother is alive, and is living in some part of Zootopia. So he still does have family.

The way Nick says it, it can easily also be interpreted that he's not on good terms with them anymore, which may or may not be the case if his mother didn't approve of his con-man ways. So that's not a huge hole, at least.
